Unless you are having an Internet connectivity issue, which sometimes happens with computer infections.
These are good tools to have ready to use at home.
I have a packed computer and it took two hours, I think an on-line scan would take a lot longer.
Although I only use Microsoft Security Essentials I think it doesn't hurt to have a third-party scanner check as well.
One can do Microsoft's scan as well, before going to bed, but as I mentioned, one needs more than one tool available.
Just like we can't expect to repair a computer if we only have one at home, you need to be able to download tools and get information from web searches if one isn't working.

If one is using Windows 10 (and probably Windows 8.1, but I don't have that handy to check) one can schedule an offline scan with Windows Security to be executed on the next restart.
