Not to ask the obvious, but do you have sound working from other applications? And is there a separate “mute” setting or volume control within that app? I’m assuming you may be using this with JAWS on, so there must be some sound happening with JAWS. I recently had a problem with one particular application that seemed to stop making sound when JAWS was operating. Can you try it with JAWS off, and then it works, you might assume it is a JAWS conflict? Sound is a strange thing on computers. There are quite a few applications that don’t like sharing audio control on the same computer at once.

Sometimes there are simply sound glitches on some machines that seem to reset with a reboot. Have you tried to reboot and launch that application immediately after rebooting? If that works, try and note what has to run afterwords to prevent Cam Video from working.
